ÖZET
Amaç: Plasenta; mükemmel bir şekilde anne-fetus arası bağlantıyı sağlayan kompleks yapısı, kök hücreden son derece zengin içeriği, inanılmaz çeşitlilik gösteren protein ve protein reseptörleri ile ilgili dinamik yapısı nedeniyle kadın doğum, rejeneratif tıp ve neonataloji alanındaki çalışmaların ilgi odağı olmaya devam etmektedir. Plasentanın farklı bölgelerindeki hücrelerin yapısal ve fonksiyonel özelliklerinin nasıl değiştiği konusundaki çalışmalar sağlıklı bir gebelik ve sonrası ile ilgili çok sayıdaki soruya cevap olacaktır. Bu araştırmada; plasenta fonksiyonlarının gerçekleşmesinde önemli ortaya konulmuş “Alfa-Fetoprotein, İnhibin, Kromogranin, Progesteron ve Östrojen reseptörlerinin plasentanın farklı bölgelerindeki lokalizasyonundaki değişimin incelenmesi amaçlanmıştır. Gereç ve Yöntemler: Normal gelişimini tamamlamış 10 insan plasenta örneğinden iki farklı lokalizasyonda hazırlanmış örnekler immunohistokimyasal işaretleme teknikleri kullanılarak işaretlenmiştir. Umbilikal korda en yakın (0 cm. uzaklıkta) lokalizasyondaki plasenta örnekleri I. grup; umbilikal korda 5 cm uzaklıktan hazırlanan plasenta örnekleri ise II. grup olarak değerlendirilerek; ilgili proteinlerin hücresel düzeydeki ekspresyonları mikroskopik olarak karşılaştırmalı şekilde değerlendirilmiştir. Bulgular: Proteinlerin plasentadaki ekspresyonu değişkenlik göstermektedir. Ancak; genel olarak umbilikal korda yakın bölgelerde tüm proteinlerin ekspresyonlarının daha düşük düzeyde olduğu gözlenmiştir. Sonuç: Hücrelerin umbilikal korddan uzaklaştıkça farklılaşmalarının artması nedeniyle, ilgili reseptörleri taşıyan daha özelleşmiş hücreler olabileceğini düşündürmektedir. Umbilikal korda en yakın yerleşimli plasental hücrelerin kök hücre özelliklerinin daha yüksek olabileceği konusundaki ilerleyen çalışmalar bu konudaki sorulara cevap olabilecektir.
ABSTRACT
Objective: Placenta remains as a focus of interest for the obstetricians, clinicians in neonatologists, and for the scientists in regenerative medicine because of its complex structure that provides perfect relatisonship between the mother and the baby, its stem cell rich component, as well as its dynamic structure related to incredible variable proteins and proteins reseptors in it. Studies that describe the differences at the location of different proteins in placenta relatod to their functions will answer many questions about healthy pregnancy and its duration. The current study aimed to examine the expression of alpha-fetoprotein, inhibin, chromogranin, estrogen receptors as well as progesteron receptors in placenta at different locations comparatively. Material and Methods: Placenta specimens from two different locations that are provided from 10 full term placenta were labeled by using immunohistochemical methods. First group specimens were located nearby of the umbilical cord and the second group contained the specimens that were 5 cm away from the umbilical cord. Expression of these five proteins at microscopic level were compared. Results: In spite of the variability of the distribution of the proteins according to their location; all protein expression levels were lower on the samples that are close to the umbilical cord. Conclusion: This suggests that the cells are differentiating when they move to the regions in ceratin distances and the cells that are closest to the umbilical cord in the placenta are reflecting more stem cell features since they are not specialized.
